I believe it means to encode the string into characters.

"mission three" should be "mission%20three" but that didn't work.

That what 'single encode' means as far as unicode is concerned.

I don't know if it's asking to encode:

mission three

- or -

"mission three"

Kind of ambiguous to me ... although every combination I've tried of both is yielding nothing.

I don't think it wants the quotes included. That is only stating to encode the string "mission three". I would leave the quotes out. The space is a different matter though because it is enclosed in the quotes, it probably should be included in the encoding scheme.
